You most likely won't need that much to get a defender, although it is just luck. The best way, in my opinion, is the kill mithril armor, because it gives exp as well as tokens. Time: Around 2-3 hours

If you have done As A First Resort, you should use the infinite run pool and do the hardest shot put. You may not need 3k tokens though.

Continue after you get your 1st rune defender... since the other you'll get will also be rune !

 

1- bank space holder (always good to take items from same location)

 

2- in case you loose 1st, you simply can go back get another one without doing all the process, just get few tokkens are there you are.
