Once you have completed the Quest, you can return to the 2nd Boss room and you will find a new Kalphite area with Kalphite Soliders/Guardians. You can go through the exit to the surface to make a new shortcut to the Kalphites and the Kalphite Queen.

 

This new Kalphite dungeon does not permit a shortcut access back to the Kalphite Queen. Returning to this area and attempting to go back through to the KQ lair, you receive a message stating 'The sides of this hole are too steep to climb up.' Additionally, upon examining it, it reads 'A cavernous hole. It looks like the passage within is too steep to climb back up.'
