I'm currently planning the layout of my house and I have a quick question with regards to room alignment. For those rooms with only 1 door, such as a portal chamber, can I choose whether the door is N,S,E or W?

You can't choose as such, but you can arrange it so it fits whichever door of whatever room to which you attach it. :-k

 

 

 

I'm not sure that makes sense, but I know what I mean and that's the second most important thing.

 

 

 

I would recommend haveing it near your front door if possible, and next to chapel (again, if possible) for future prayer training. I have mine upstairs which is quite slow. If I ever get insanely rich, I'll build a new house.

So for example if I wanted a portal chamber immediately to the right of my garned, how would I build the portal chamber to ensure the door is on the West side?

I think from memory there is an option to rotate the room when you first create it

 

 

 

I know it can be done as I have two portal rooms, each with a door in an opposite direction.

IIRC (I haven't tested this. I will in a bit) the door must be facing where you're creating the it via the door hotspot. (I.E. if the door you're building it off faces west, the portal door MUST face east, if the door spot faces north, the portal door MUST face south. You can't rotate one-door rooms to my knowledge.
