Do you need the right defence for each gloves or are the no defence requirements? ie 20 defence for mithril gloves.

No there are not any defence requirements for these gloves.

The reason why the gloves don't have any defence requirements is because they are just gloves with good stats. The whole mith glove, dragon glove, rune glove, etc thing was created by ppl to differentiate between the gloves.
