I currently have the Adamant & Rune defenders, do I have to keep the Adamant one in order to obtain more Rune defenders ? Or I can just throw away the Addy one and keep the Rune one and keep getting more Rune defenders ?

You can use the Rune Defender to get more Rune Defenders. I'd get a spare before you completely ditch the addy one though.

I'd keep the addy one just in case you ever lose your defenders. But you can just throw it away.
