You need to have the Last.fm client downloaded to have it "scrobble" (pick up) your plays from iTunes, WMP, Winamp, whichever you choose. Just type in "download last.fm" into google and you should find it easily. And if you have an iPod, it can scrobble your plays from that every time you connect it to your computer. :)
